Scientists find missing link between players in the epigenetic code

ScienceDaily (Sep. 30, 2012) ? Over the last two decades, scientists have come to understand that the genetic code held within DNA represents only part of the blueprint of life. The rest comes from specific patterns of chemical tags that overlay the DNA structure, determining how tightly the DNA is packaged and how accessible certain genes are to be switched on or off.

As researchers have uncovered more and more of these "epigenetic" tags, they have begun to wonder how they are all connected. Now, research from the University of North Carolina School of Medicine has established the first link between the two most fundamental epigenetic tags -- histone modification and DNA methylation -- in humans.

The study, which was published Sept. 30, 2012 by the journal Nature Structural & Molecular Biology, implicates a protein called UHRF1 in the maintenance of these epigenetic tags. Because the protein has been found to be defective in cancer, the finding could help scientists understand not only how microscopic chemical changes can ultimately affect the epigenetic landscape but also give clues to the underlying causes of disease and cancer.

"There's always been the suspicion that regions marked by DNA methylation might be connected to other epigenetic tags like histone modifications, and that has even been shown to be true in model organisms like fungus and plants," said senior study author Brian Strahl, PhD, associate professor of biochemistry and biophysics in the UNC School of Medicine and a member of UNC Lineberger Comprehensive Cancer Center. "But no one has been able to make that leap in human cells. It's been controversial in terms of whether or not there's really a connection. We have shown there is."

Strahl, along with his postdoctoral fellow Scott Rothbart, honed in on this discovery by using a highly sophisticated technique developed in his lab known as next generation peptide arrays. First the Strahl lab generated specific types of histone modifications and dotted them on tiny glass slides called "arrays." They then used these "arrays" to see how histone modifications affected the docking of different proteins. One protein -- UHRF1 -- stood out because it bound a specific histone modification (lysine 9 methylation on histone H3) in cases where others could not.

Strahl and his colleagues focused the rest of their experiments on understanding the role of UHRF1 binding to this histone modification. They found that while other proteins that dock on this epigenetic tag are ejected during a specific phase of the cell cycle, mitosis, UHRF1 sticks around. Importantly, the protein's association with histones throughout the cell cycle appears to be critical to maintaining another epigenetic tag called DNA methylation. The result was surprising because researchers had previously believed that the maintenance of DNA methylation occurred exclusively during a single step of the cell cycle called DNA replication.

"This role of UHRF1 outside of DNA replication is certainly unexpected, but I think it is just another way of making sure we don't lose information about our epigenetic landscape," said Strahl.

Former Zeeland attorney to plead in $900,000 ... - Estate of Denial

GRAND RAPIDS, MI ? Former Zeeland attorney Kenneth Hoesch has agreed to plead guilty in a scheme to steal nearly $900,000 in clients? estate planning trusts.

Hoesch, recently found competent to stand trial, has agreed to plead guilty to charges of mail fraud, a 20-year felony, and making a false statement on a tax return, a three-year felony.

The government today filed felony information outlining the plea agreement.

Hoesch, while a licensed attorney, set up estate planning trusts for clients.

?In truth and fact, defendant Hoesch engaged in a fraudulent scheme to deprive his law clients and their intended beneficiaries of money, funds and similar property rights by the commission of a mail fraud scheme ?,? Assistant U.S. Attorney Michael MacDonald wrote in court documents.

He said Hoesch took clients? assets, and did not place them in trust accounts, or placed them in trust accounts only to withdraw funds for his own use. He also moved clients? funds to conceal the theft, and issued false reports, statements and correspondence.

Hoesch also concealed the fraud from his former law partners.

Both sides in the case agreed that restitution should be $889,398.

The agreement also requires Hoesch to pay restitution of $835,000 to Grand River Bank. He had provided false and misleading information that resulted in the bank issuing two loans to him, documents showed.

Those loans were secured by real estate.

The government also said Hoesch took $700,000 from a client from 1997 to 2002, which is outside of the statute of limitations.

Both that case and the alleged bank fraud will be used in calculating losses for federal advisory sentencing guidelines.

He is scheduled to plead on Wednesday in U.S. District Court in Grand Rapids.

Max Scherzer was skipped in the rotation this week due to a nagging shoulder issue, but he could end up playing a critical role for the Tigers before the end of the regular season.

US immigration chief: Same-sex ties are family ties

Same-sex couples will be considered ?family relationships? in immigration proceedings, according to Department of Homeland Security Secretary Janet Napolitano, a move that could help stem the deportation of those in gay or lesbian binational relationships.

Close family ties to the United States are a factor considered by authorities in deportation cases, and gay and lesbian advocates have long argued for same-sex couples to have the same immigration rights as opposite-sex couples.

?In an effort to make clear the definition of the phrase ?family relationships,? I have directed ICE to disseminate written guidance to the field that the interpretation of the phrase ?family relationships? includes long-term, same-sex partners,? Napolitano said in a letter.

Eight-four members of Congress signed a joint letter to Napolitano on July 31 asking for her to put into writing an order to prevent the deportation and separation of immigrants from their American citizen same-sex partners.

?One of those who penned the letter, U.S. Congressman Michael Honda of California, said Napolitano?s response, which he received Thursday night, heralded ?promising news.?

?In the wake of this important victory, we must take a step forward and continue the fight for immigration reform. Current immigration laws are tearing families apart and separating American citizens from their loves ones,? he said in a statement.??No one should have to choose between their spouse and their country, and no family should be left out of the immigration system.?

There are an estimated 36,000 binational gay couples in the U.S. Two such couples have brought lawsuits challenging the Defense of Marriage Act, a U.S. law passed in 1996 that bars federal recognition of same-sex marriages and thereby denies various benefits given to heterosexual couples, such as the right to immigrate.

Rachel B. Tiven, executive director of Immigration Equality, called the announcement a ?huge step forward.?

?Until now, LGBT families and their lawyers had nothing to rely on but an oral promise that prosecutorial discretion would include all families. Today, DHS has responded to Congress and made that promise real. The Administration?s written guidance will help families facing separation and the field officers who are reviewing their cases,? she said in a statement.

Tiven was referring to the prosecutorial discretion laid out in June 2011, when ICE Director John Morton issued a memo requiring staff? to consider the circumstances presented in individual deportation cases, such as whether the person has close family ties to the U.S.

When will it ever go into production, and how much will it cost if it does? The company still isn't saying, but if you have to ask...

Scaling up polymer blobs

ScienceDaily (Sep. 27, 2012) ? Scientists use simulations to test the limits of their object of study -- in this case thin films of polymers -- to extremes of scale. In a study about to be published in EPJ E, Nava Schulmann, a researcher at Strasbourg University, France, and colleagues use a well-known model capable of providing information on heat and mechanical energy exchange between these polymer chains. They found that polymer blends confined to ultrathin two-dimensional films displayed enhanced compatibility. This was made possible by simulations using a fairly standard model, which is simple enough to allow the efficient computation of dense large-chain systems.

The authors focused on making simulations of self-avoiding and highly flexible polymer chains without chain intersections. To do so, they varied the level of polymer density, as well as their chain length, while using numerical methods to arrive at a universal view of polymer behaviour.

Thanks to molecular dynamics and so-called Monte Carlo simulations, they confirmed that such polymers adopt a scaling behaviour following a power law as a function of density and chain length. This scaling behaviour applies, for example, to polymer pressure and, hence, polymer compressibility. French Nobel laureate Pierre-Giles de Gennes predicted this property in his so-called blob picture approach. Accordingly, a polymer chain is akin to a succession of blobs, like beads in a necklace.

Schulmann and colleagues focused on a regime relevant for applications, referred to as a semi-dilute regime. There, scaling occurs more universally as long as the initial blob size is well defined. Understanding the limit of a system of long chains can currently only be realised in simulations of simplified models. However, the authors hope their findings will facilitate the work of polymer experimentalists.?

I would like to join but, I was wondering what kind of inventions of magitech are there. Are there magical computers? Magical Surveillance Sensors. I would like to be sort of magical hacker. In a sense it is sort of like a Seer in a way. Just using magical technology to make it happen.

Is the magitech on level with computer and internet being quite common. Can people communicate via special magitech earpieces?

If not I'll have to come up with something else. I have another idea, but the hacker is my favorite so far.

Noida farmers want denotification of City Center ... - Real Estate India

NOIDA: Villagers of Noida seem unrelenting a day after the Supreme Court issued a notice to UP government over the petition of 94 Noida farmers from Nithari, Morna and Chaura villages accusing the Authority of misusing its powers by acquiring land in 1976 and letting it lie idle for 34 years. The farmers want denotification of the land that was allotted to Wave Infratech in March last year for the ambitious City Center project.

Noida Authority refused to comment on the implication of the order before it receives a copy of the SC notice even as work on the site is currently on. Sources said that while the Authority will submit a ?suitable reply to the court once it receives the copy of the notice. The notice by itself does not imply that work at the City Center site needs to be halted.?

?We can comment on the matter only after we deliberate on the matter with our legal officers. However, the state has never been against the farmers and anyone who has a problem can approach the Authority for redressal,? Authority officials said. The Authority also refused to comment on why the 152 acre plot was kept idle for 34 years.

Through a written statement, the executive director of Wave Infratech, R K Jain, said, ?Wave Infratech acquired the 152 acre of land in 2011 through fair competitive bidding from Noida authorities by outbidding other real estate developers with the winning bid of Rs 6,500 crore. We never purchased this land from the farmers directly, so the said issue is between the farmers and the state government.?

?The Authority did not give us a fair hearing at the time of acquiring our land. We feel cheated and we have told the court we must be given our land back,? said Dinesh Sharma of Nithari village.

Meanwhile, another group of greater Noida farmers is now protesting against another Wave Infratech project, the ?Wave Hi-tech city? that falls partly in Greater Noida and partly in Ghaziabad. These farmers are demanding compensation at par with Noida Extension villagers and have called for a mahapanchayat on Wednesday in Dujana village.

MANILA, Philippines ? A new $35-million credit facility which is a key component of a broader US government effort to promote economic growth in the Philippines, will provide increased access to capital for the country?s small and medium enterprises (SMEs).

The credit facility is a key component under the US-Philippines Partnership for Growth.

Launched last November by US Secretary of State Hillary Clinton and Foreign Affairs Secretary Albert del Rosario, the Partnership for Growth will help the country move to a higher, sustained and more inclusive growth trajectory in line with other high-performing economies.

US Ambassador Harry Thomas Jr. and US Agency for International Development (USAID/Philippines) mission director Gloria Steele launched the eight-year, $35-million credit facility together with?Bank?of?the?Philippine?Islands?(BPI) president and chief executive officer Aurelio Montinola III and BPI Family Savings Bank (BFSB) president Jose Teodoro Limcaoco.

?This credit facility will help expand access to capital to allow Filipino entrepreneurs improve and develop their businesses. More importantly, increased access to credit will encourage the creation of more businesses, resulting in more employment opportunities and increased income for Filipinos,? Thomas said.

The facility will encourage lending to SMEs in three cities outside Metro Manila ? Batangas, Iloilo and Cagayan de Oro ? that are the focus of USAID?s Cities Development Initiative (CDI).

An integral part of the Partnership for Growth, the CDI aims to promote growth in cities identified for their growth potential, and to unlock and maximize the country?s potential.

?USAID?s partnership with BPI and BPI Family Savings Bank aims to stimulate targeted domestic commercial investment and increase the number of start-up businesses in the CDI cities and surrounding areas. With an eight-year coverage and five-year enrollment period, the credit facility will help reduce SME collateral requirements and allow entrepreneurs to explore new markets,? Steele said.

Montinola said BPI and BPI Family Savings Bank have long been engaged in endeavors that would help SMEs bring progress to the whole economy.

Is "Illegal Immigrant" "Inflammatory, Imprecise, And ... Inaccurate"?
That's what Jose Antonia Vargas writes in TIME:

Add that to the list of questions I am repeatedly asked since publicly disclosing my undocumented immigrant status in the summer of 2011. Calling undocumented people "illegal immigrants" -- or worse, "illegal aliens," as Mitt Romney did in front of a largely Latino audience last week -- has become such standard practice for politicians and the media, from Bill O'Reilly to the New York Times, that people of all political persuasions do not think twice about doing it, too.

But describing an immigrant as "illegal" is legally inaccurate. Being in the country without proper documents is a civil offense, not a criminal one. (Underscoring this reality, Justice Anthony Kennedy wrote for the majority opinion on SB 1070, Arizona's controversial immigration law: "As a general rule, it is not a crime for a movable alien to remain in the United States.") In a country that believes in due process of the law, calling an immigrant "illegal" is akin to calling a defendant awaiting trial a "criminal." The term "illegal" is also imprecise. For many undocumented people -- there are 11 million in the U.S. and most have immediate family members who are American citizens, either by birth or naturalization -- their immigration status is fluid and, depending on individual circumstances, can be adjusted.

When journalists, who are supposed to seek neutrality and fairness, use the term, they are politicizing an already political issue. (How can using "illegal immigrant" be considered neutral, for example, when Republican strategist Frank Luntz encouraged using term in a 2005 memo to tie undocumented people with criminality?) And the term dehumanizes and marginalizes the people it seeks to describe. Think of it this way: In what other contexts do we call someone illegal? If someone is driving a car at 14, we say "underage driver," not "illegal" driver." If someone is driving under influence, we call them a "drunk driver," not an "illegal driver." Put another way, how would you feel if you -- or your family members or friends -- were referred to as "illegal"?

Well, since those friends of mine who emigrated here from other countries went through hell to do it legally, having to go through long and awful processes in a couple cases, that would really suck.

I looked up "illegal" in the Apple dictionary (on my computer):

illegal |i(l)?l?g?l| adjective

contrary to or forbidden by law, esp. criminal law: illegal drugs.

noun
an illegal immigrant.

And yes, it really does give that example.

It is illegal to cross our borders and enter this country without permission. Thus, it seems correct to call someone who does so an "illegal immigrant."

This differentiates them from people who follow the law -- i.e., legal immigrants.

We don't call people illegal hijackers, illegal home-invaders or illegal muggers because there are no legal hijackers, home-invaders or muggers.

Also, a commenter at TIME, Talendria, points out that the only violation of our laws usually isn't just at the border crossing:

When people use the term "illegal," they're typically not referring to a visitor who overstayed his visa accidentally or on purpose. They're referring to people who willfully engaged in illicit activity to sneak into the United States, obtain a job, and benefit from social services because they knew they weren't eligible under the existing immigration laws.

While I'm not an expert in criminal law, I'm guessing all of the following activities are illegal to some extent:
--forging documents
--buying forged documents
--using forged documents to obtain a job
--driving without a valid license
--driving without insurance

There are additional activities which may not be strictly illegal but are still harmful:
--enrolling non-citizens in public schools
--taking advantage of social services meant for citizens (food, housing, health care)
--taxing the legal system (police call-outs, court appearances, prison)

I think people cling to the term "illegal" even though they know it sounds uncharitable because they're offended by the blatant disregard for our laws and the apparent lack of concern for the social and economic consequences of breaking those laws.

I personally would love to find a way to resolve this problem to everyone's satisfaction, but you've drawn your line in the sand so far to the left that it's difficult to find any common ground.

P.S. It's a felony in Mexico to be an illegal immigrant. (I don't speak Spanish, but I'm guessing they don't use a frilly, P.C. term for it, either.)

Martin's HR in 10th lifts hot Yanks over A's

NEW YORK (AP) - When Rafael Soriano gave up a tying homer in the ninth inning to spoil CC Sabathia's dominant start, the Yankees didn't get down. They knew what they needed to do to keep their AL East lead.

Russell Martin led off the bottom of the 10th inning with a homer off Sean Doolittle, and New York maintained its one-game division lead by beating the Oakland Athletics 2-1 Friday night for its sixth straight win.

"Good teams get each other's back," Martin said.

Sabathia pitched encouraging three-hit ball over eight innings to help the Yankees stay ahead of Baltimore, which beat Boston 4-2 for its fifth straight win. The A's, losers of four of five, dropped a game back of the Orioles for the top wild-card spot. Oakland started the day with a 4 1/4-game lead over the Angels.

Pinch-hitter Brandon Moss hit a towering home run to right field with one out in the ninth off Soriano to tie it 1-all and stun the boisterous crowd of 40,759 into silence. It was Soriano's fourth blown save in 46 chances.

"His arm may have been a little dead today," Joe Girardi said of Soriano, who saved both games of a doubleheader Wednesday against Toronto.

But Soriano disagreed. He said he had "normal soreness" and would go through his routine Saturday before letting his manager know if he will be available.

Curtis Granderson had a sacrifice fly in the fourth inning off Jarrod Parker and Ichiro Suzuki continued his torrid pace with two more hits - one getting caught in Parker's jersey.

The Yankees lost four-straight one-run games in Oakland in mid-July that began a summer swoon in which they blew a 10-game division lead. Martin's second walk-off homer this season gave the Yankees their 11th win in 15 games.

"You can feel that playoff weather coming," Martin said. "I like it. I enjoy it."

David Robertson (2-7) pitched a perfect 10th for the victory. Doolittle (1-1) pitched the ninth before giving up Martin's homer on an 0-1 pitch.

The catcher flinged his helmet almost all the way to first base as he approached home then was engulfed by his teammates as he stepped on home plate.

"Right off the bat I knew it was gone," Sabathia said. "He called a great game tonight and kept the guys off-balance and it worked out."

Parker was in line for a loss before Moss' homer despite matching Sabathia for eight innings. He gave up six hits and struck out seven in his first start at Yankee Stadium.

"It's really frustrating," Doolittle said. "He did such an unbelievable job going toe-to-toe with Sabathia."

Sabathia's performance had become one of the biggest question marks in the Yankees rotation the past several weeks. The big lefty was 0-3 in his last four starts and he gave up leads in each outing. After two stints on the disabled list, his velocity was down but he insisted he was OK. Girardi said Sabathia was struggling with the command of his sinker and changeup.

He quickly put that to rest by striking out the side in the first with three breaking pitches. His velocity went up as the innings went by, ending the sixth by blowing a 96 mph a fastball past Jonny Gomes.

"I was just trying not to overthrow. I felt pretty good coming out of the bullpen," Sabathia said. "I just wanted to make sure I kept my delivery together and make sure I commanded the ball. The velocity will come and go so it's up to me to make pitches."

After a walk to Gomes with one out in the first, Sabathia retired 14 in a row before Stephen Drew singled leading off the sixth.

Sabathia gave up another hit to Drew with one out in the eighth. With two outs, Collin Cowgill beat out an infield single and Sabathia grazed Gomes with a pitch to load the bases.

Pitching coach Larry Rothschild visited the mound as Robertson warmed up in the bullpen. But Sabathia remained and got J.J. Reddick to fly out to left field on the first pitch of the at-bat, ending the inning.

The A's lead the majors in striking out and they fanned 11 times while walking twice against Sabathia.

"You want him feeling good about himself, confident," Martin said. "I think he definitely needed this start for himself."

Any questions about Parker being nervous in his first start in the Bronx were dismissed right away with an eight pitch inning. He didn't give up a hit until he lost Suzuki's bouncer back to the mound with one out in the third inning in his jersey. Parker grimaced in frustration as he failed to pull the ball out of his green and gold top.

The hit was Suzuki's 10th in his last 13 at-bats. Suzuki also had a hit in the sixth but was thrown out trying to stretch it to a double.

The Yankees scored in the fourth when Nick Swisher led off with a more traditional hit, a lined single to right field. Alex Rodriguez followed with a sharp single to center field. Granderson then hit a sacrifice fly.

Derek Jeter extended his hitting streak to a season high-tying streak of 15 games with a sixth-inning single.
